The Plasmid DNA Isolation Kit delivers rapid and reliable purification of high-quality plasmid DNA, accommodating both high- and low-copy-number plasmids of various sizes. The optimized silica-based membrane chemistry ensures efficient recovery of pure plasmid DNA suitable for all molecular biology workflows, including cloning, PCR, sequencing, and transfection. Depending on the plasmid copy number and growth conditions, yield and culture volume may vary, but consistently high-quality DNA is obtained for all downstream applications.

The kit employs the alkaline lysis method followed by selective binding of plasmid DNA to a silica membrane under high-salt conditions. After cell lysis and neutralization, chromosomal DNA and debris are removed by centrifugation. The clarified lysate is loaded onto the spin column, where plasmid DNA binds efficiently. Following washing steps to eliminate impurities, pure plasmid DNA is eluted in a low-salt buffer such as 10 mM Tris-HCl (pH 8.0).
